چکیده

Water-soluble dinuclear Ru-arene complexes were synthesized and found to exert promising cytotoxic effects in human cancer cells, which could be increased to an IC50 of 0.29 μM by increasing the spacer length between the metal centers. Cytotoxicity could be correlated with lipophilicity (log P Values) and water solubility. The most potent dinuclear compound, 1,12bis{chlorido[3-(oxo-κO)-2-methyl-4-pyridinonato-κO4](η-pisopropyltoluene)ruthenium}dodecane, is at least 2-3 orders of magnitude more actiVe than the mononuclear analogue chlorido[3-(oxo-κO)-2-methyl-4-pyronato-κO4](η-p-isopropyltoluene)ruthenium.

Influence of the Arene Ligand, the Number and Type of Metal Centers, and the Leaving Group on the in Vitro Antitumor Activity of Polynuclear Organometallic Compounds

Dinuclear ruthenium complexes were shown to exhibit strong antiproliferative properties in human tumor cell lines. In order to extend the structure-activity relationships (SARs), a series of new Ru(arene)X complexes (X = Cl, Br, I) linked by pyridinone-based spacers were synthesized and assayed for their in vitro antineoplastic effect. The SARs were established in terms of the arene ligand, the...

Transferring the concept of multinuclearity to ruthenium complexes for improvement of anticancer activity.

Multinuclear platinum anticancer complexes are a proven option to overcome resistance of established anticancer compounds. Transferring this concept to ruthenium complexes led to the synthesis of dinuclear Ru(II)-arene compounds containing a bis(pyridinone)alkane ligand linker. A pronounced influence of the spacer length on the in vitro anticancer activity was found, which is correlated to the ...

Mono- and dinuclear (η-arene) ruthenium(II) benzaldehyde thiosemicarbazone complexes: Synthesis, characterization and cytotoxicity

Anticancer drugs A series of monoand dinuclear (η-arene) ruthenium(II) complexes were prepared by reaction of thiosemicarbazone ligands derived from benzaldehyde and ruthenium(II) precursors of the general formula [Ru(η-arene)(μ-Cl)Cl]2, where arene=p-PrC6H4Me or C6H5C3H6COOH. These complexes were characterized by NMR and IR spectroscopy, ESI-mass spectrometry and elemental analysis. The molecu...

Anticancer activity of new organo-ruthenium, rhodium and iridium complexes containing the 2-(pyridine-2-yl)thiazole N,N-chelating ligand

The dinuclear dichloro complexes [(g-arene)2Ru2(l-Cl)2Cl2] and [(g-C5Me5)2M2(l-Cl)2Cl2] react with 2-(pyridine-2-yl)thiazole (pyTz) to afford the cationic complexes [(g-arene)Ru(pyTz)Cl] (arene = C6H6 1, p-PrC6H4Me 2 or C6Me6 3) and [(g-C5Me5)M(pyTz)Cl] (M = Rh 4 or Ir 5), isolated as the chloride salts. The reaction of 2 and 3with SnCl2 leads to the dinuclear heterometallic trichlorostannyl de...

Arene ruthenium complexes as anticancer agents.

Neutral or cationic arene ruthenium complexes providing both hydrophilic as well as hydrophobic properties due to the robustness of the ruthenium-arene unit hold a high potential for the development of metal-based anticancer drugs.
